Brief CV:
Professor Hambley has studied and worked at the Universities of Western
Australia and Adelaide, ANU, The University of Sydney, Emory University
and Birkbeck College, University of London. He was the recipient of the
Edgeworth David Medal in 1989 for contributions to Science by an
Australian under the age of 35. He received the "Supervisor of the Year"
award at the University of Sydney in 1997 and an Excellence in Teaching
Award for Postgraduate Supervision in 1998.
Over the last 17 years Prof. Hambley has developed a reputation in the study of the mechanism of action of Pt based anticancer drugs and their interactions with biomolecules. He has been invited to present the results of this work at twelve major international meetings since 1999 including the 8 and 9th International Symposia on Platinum and Other Metal Coordination Compounds in Cancer Chemotherapy (ISPCC-8 and -9), the 9th International Conference on Bioinorganic Chemistry (ICBIC-99), the American Chemical Society meeting in San Francisco in March 2000, a conference celebrating the work of Mendel in Brno, Czech Republic in July 2000, the PacificChem conference in Hawaii in December 2000, ICBIC-2001 (Florence, August, 2001), Frontiers in Chemical Design and Synthesis (SICC2, Singapore, December 2001) and Eurobic 2004 (Germany, September, 2004). He was invited to chair the nucleic acids section at the inaugural Metals in Medicine Gordon Conference (Boston, July 2002).
Prof. Hambley has published 3 books, 12 reviews and more than 400 refereed journal papers, including four recent reviews of the relationship between the structure of Pt anticancer agents and their toxicity and anticancer activity and on the modelling of Pt/DNA interactions. He has had four recent invitations to produce book chapters, three to be published in 2003 and one in 2004. At the end of 2001, his publications had received 3205 citations excluding self-citations, an increase of 309 (10.7%) in the previous twelve months. Based on citations in 1981-1997, he ranks as the 528th most cited chemistry author worldwide which places him in the top 0.1%. He is a member of the Editorial Boards of the Journal of Inorganic Biochemistry and the Journal of Biological Inorganic Chemistry. He was the initiator and a Co-guest Editor of a special issue of this journal that arose from the 8th International Symposium on Platinum and Other Metal Coordination Compounds in Cancer Chemotherapy.
Brief summary of expertise and significant research achievements:
Professor Hambley (CI) has contributed to a number of areas including
molecular modelling, crystallography and medicinal inorganic chemistry.
His most recent contributions have been in the area of medicinal inorganic
chemistry and in particular Pt anticancer drugs.
Prof. Hambley was among the first to employ theoretical modelling to investigate Pt/DNA interactions and the first to show how these methods could be used to rationally design complexes to control their interactions with DNA. This led to the design of complexes that are as effective as cisplatin in interstrand cross-linking DNA, but are 50 fold less active, providing strong evidence that interstrand adducts are not primarily responsible for anticancer activity in these complexes. It has also led to the design of a series of chiral complexes that have greatly improved the understanding of the factors that control the binding of Pt to DNA. With Prof. Berners-Price, he was the first to obtain experimental confirmation that Pt/DNA adduct distributions are determined at the level of monofunctional adduct formation. Together, this work led to an invitation to write a Perspective for Dalton Transactions on the factors influencing platinum(II) binding to DNA.
More recently, Prof. Hambley has made major contributions to the understanding of the chemistry and biochemistry of platinum(IV) complexes. He was the first to investigate the relationship between the reduction potentials of Pt(IV) complexes and their biological activity, now a rapidly developing area. He has also developed independently, and with others, methods for controlling the solubility and lipophilicity of platinum(IV) complexes. This work has resulted in an association with IDT Australia aimed at taking a number of novel platinum(IV) complexes into the clinic.
Most recently, Prof. Hambey and his group have been the first to develop synchrotron radiation based methods for the in situ determination of oxidation states of platinum in biological environments and for reliably determining platinum distributions in these environments. Prof. Hambley has also investigated cobalt complexes as delivery agents for matrix metalloproteinase inhibitors and used XANES to determine oxidation states of the cobalt complexes in solution. This work is recognised as being world leading and has led to an invitation to use a new beam line at the Advanced Photon Source in Chicago and to be a speaker at the APS Biological Applications of Hard X-ray Microscopy Workshop.
